Pavel Nedved set up both goals for his fellow midfielders as Juve end Sampdoria's seven-match unbeaten run.
This was an emotional moment for Marcello Lippi, as the Juventus boss spent much of his playing career with the Sampdoria jersey on his back.
The Blucerchiati have been out of the top flight for several seasons, but made a remarkable comeback and are currently fighting for UEFA Cup spots.
Fabio Bazzani returned from injury to renew his partnership with the on form Francesco Flachi, but Cristiano Doni pulled out with a muscular problem just before the match.
Juve have now waved goodbye to Edgar Davids, who is set to complete his switch to Barcelona next week, while Alessio Tacchinardi and Lilian Thuram are out injured. Enzo Maresca therefore retained his spot in midfield.
The Bianconeri had the better of the early play and would create fine moves from midfield, but David Trezeguet ballooned over the bar from ten yards.
The deadlock was eventually broken on 23 minutes when Pavel Nedved's sweeping cross found a Mauro German Camoranesi diving header at the back post.
The Sampdoria players were furious, as they maintained that Nedved had taken the ball off Cristian Zenoni through dangerous play.
Set-piece specialist Sergio Volpi went close to an equaliser when his free kick from 20 metres dipped inches over the bar.
There was a good opportunity to make it two for Juve on the half-hour mark, but Francesco Antonioli intercepted an Alessandro Del Piero cutback before it could reach the unmarked Trezeguet.
Massimo Donati slammed a volley wide of the far post as Samp tried to fight back.
The start of the second half was marred by an incident that will cost Sampdoria dearly. Someone in the crowd behind Gianluigi Buffon's goal threw a spray can that struck him on the back of the head. The goalkeeper was medicated for a cut, but was able to carry on.
Del Piero frankly should have doubled Juve's lead soon after, but somehow failed to tap into an empty net from less than a yard following another Nedved centre.
Samp saw penalty appeals for a Ciro Ferrara handling offence waved away, as the defender was trying to protect his face from Donati's cross.
They increased the pressure and drew level on 56 minutes with that man Francesco Flachi yet again. He was left unmarked for a free header on Volpi's smart centre.
However, it did not last long, as Juventus restored their lead soon after. Nedved was again involved with an intelligent backheel for Antonio Conte's angled drive.
Nedved set up both Juve goals, but was unable to score his own when firing over from close range.
Samp gave new signing Giacomo Cipriani his debut in the final minutes, but despite a spirited attempt to equalise and a difficult late Buffon save on Flachi's close-range header, their unbeaten run ends after seven games.
